Cholesterol began to be accepted after the Second World War as a significant cause of atherosclerosis and associated conditions, such as coronary heart disease (CHD). This Witness Seminar, chaired by Professor Michael Oliver, discussed the increasing understanding of its basic science. Early epidemiological studies demonstrated the relationship between excess saturated fat consumption and elevated levels of cholesterol, although cholesterol alone did not explain all population differences among gender, class, or heritage. Work on lipoprotein metabolism pointed to the establishment of hyphercholesterolaemia as a major risk factor for CHD, culminating in the development of cholesterol-lowering drugs, particularly the successful statins, available in the UK from the early 1980s, and confirmed by randomized controlled trials. The role of diet in heart disease has remained controversial in the UK, notwithstanding the food industry’s introduction of functional foods such as cholesterol-lowering margarine. Later, recommended limits on the composition of dietary fat were agreed, and although extreme diets could reduce cholesterol, patient conformity remains difficult. Appendices on the diet-heart hypothesis and the development of lovastatin compliment the transcript. We explored plasma biomarkers of dairy fat intake, and evaluated whether higher concentrations of these biomarkers are associated with a reduced risk of fatal CHD.;We used cohort analysis in 10,555 men and women in the Campaign against Cancer and Heart Disease Study (1989-2002). Dairy intake was estimated using a Block food frequency questionnaire. We used a case-cohort design in 517 fatal CHD cases and 515 subcohort controls randomly selected from the baseline total cohort. Fatty acids were measured in frozen plasma collected at baseline. We used Cox and weighted Cox proportional hazard models to estimate hazard ratios of coronary heart disease.;The main findings are: intake of total dairy products may not be associated with an increased CHD mortality; intake of high-fat dairy products may reduce the risk of fatal CHD; and intake of low-fat dairy products may increase the risk of CHD death. Plasma contents of pentadecanoic acid and conjugated linoleic acid may be used as biomarkers of dairy fat intake. Dairy fat intake is not associated with an increased risk of fatal CHD. This web-site was targeted to two audiences: RDs and a self care program for the military beneficiaries. This self care program was created using the framework of Rosenstock’s Expanded Health Belief Model (EHBM).;The effectiveness of a web-based self care program was evaluated using a randomized 4-month study with participants either in the web-based group (n=17) or the usual care (n=13) at a U.S. Army hospital. Data were collected at baseline, two months and four months. Variables measured were anthropometric, blood pressure, lipid profile, fasting glucose, C-reactive protein, nutrient intake, physical activity, and EHBM constructs. Data were analyzed using analysis of covariance and using baseline means to adjust the two and four month data. Results indicated a significant reduction of total blood cholesterol, LDL-cholesterol, predicted body fat percent, and estimated body mass index (P<0.05) in the web-based group. The usual group demonstrated a significant increase in self-efficacy score at month four (p<0.05).;Significant within group changes for both groups were demonstrated for the reduction in waist circumference and serum triglycerides (p<0.05). Stearoyl-CoA desaturase (SCD) is the rate-limiting enzyme responsible for the biosynthesis of monounsaturated fatty acids (MUFAs). MUFAs are important substrates for the synthesis of various kinds of lipids that are essential to many key biological functions. Therefore, determining the regulation of SCD is critical to the understanding of its role in different biological pathways. SCD1 isoform is primarily regulated at the level of gene transcription by many transcription factors, and identification of certain transcription factors, such as sterol regulatory element-binding protein-1c (SREBP-1c), has implicated a role of SCD1 in lipid metabolism. Subsequent studies with SCD1 deficient (SCD1-/-) mice further demonstrated an important role for SCD1 in lipid metabolism. To further explore the role of SCD1 in lipid metabolism, transcriptional regulation of SCD1 gene by the nuclear receptor liver X receptor (LXR) was investigated. LXRs are important regulators of cholesterol and lipid homeostasis. Regulation of SCD1 gene by LXR was presumed to be primarily mediated by the master regulator of fatty acid synthesis SREBP-1c. However, here we demonstrated that SCD1 gene is also regulated by direct transcriptional activation by LXR through an LXR response element identified in the 5′ promoter region of SCD1 gene. SCD1-/- mice were then employed to study the effect of SCD1 deficiency on LXR agonist T0901317-induced accumulation of hepatic and plasma triglyceride. Upon T0901317-mediated LXR activation, SCD1-/- mice had reduced hepatic triglyceride accumulation and were markedly protected against plasma accumulation of triglyceride-rich very-low density lipoproteins (VLDLs). Further studies demonstrated that SCD1 deficiency did not reduce T0901317-induced secretion of large, triglyceride-rich VLDLs. However, SCD1 deficiency did modulate the MUFA and saturated fatty acid (SFA) content of the triglycerides packaged onto the VLDLs, resulting in the attenuation of the

In multiple linear regression models, police officers had an increased mean common carotid IMT (0.031mm, p=0.02), mean maximum carotid IMT (0.043mm, p=0.02), and decreased % FMD (-0.750%, p=0.12) compared to the control sample, after adjustment for age, gender, and CVD risk factors [Body Mass Index (BMI), education, race, hypercholesterolemia, hypertension, diabetes, smoking, alcohol consumption, physical activity, and Center for Epidemiologic Studies Depression (CES-D) score]. In order to reduce healthy worker bias, an additional analysis was conducted with a restricted sample that only included employed controls. Police officers still exhibited significantly increased structural and functional subclinical CVD compared to controls, even after adjustment for all variables previously listed.;Discussion.
